Basically it's a joke from evangelion. The character shown is normally, to put it politely, a little bitch who cries about everything. However in this one scene he just stares at the other person with that pissed off demeanor while holding that mug. It was just such a stark contrast to his normal character that it became a big joke on tumblr.

Also the mug he's holding has a picture of him holding a mug on it and all of this is on a mug.
